As one of the newly emerged super-resolution microscopy techniques, Stochastical Optical Reconstruction Microscopy (STORM) / Photoactivated Localization Microscopy (PALM) has pushed the resolution of optical microscopy beyond the classic diffraction limit by more than an order of magnitude. Our ultimate goal is to use this method to visualize protein-scale structures in cells and even living tissue. To achieve this goal, we have developed compressed-sensing-based image processing methods for faster temporal resolution, interferometric single-molecule localization to get higher spatial resolution, and active stage stabilization and aberration correction methods to image deeper into a tissue sample. We have used super-resolution microscopy to study structure of protein complexes and protein interactions controlling the development of motor neurons in Drosophila embryo.
